Causal inference from observational data is challenging due to confounding and reverse causation. Mendelian randomization (MR) is a method that uses genetic variants as instrumental variables to infer the causal effect of a modifiable exposure on an outcome in the presence of unmeasured confounding. In this talk, I will introduce the basic concepts of MR and discuss methodological challenges such as weak instruments and heterogeneous causal mechanisms. I will also present empirical studies that apply MR and mediation analysis using large-scale biomedical databases, including the Taiwan Biobank, the UK Biobank, and the Genotype-Tissue Expression project, to investigate causal mechanisms underlying various diseases and potential drug targets.
